Một ngày nọ, mặt trăng bất thình lình bị một sinh vật siêu việt thổi bay 70%. Sinh vật ấy đã tự nhận mình là thủ phạm, và tuyên bố sẽ thổi bay toàn bộ Trái đất vào tháng Ba năm sau. Không cường quốc nào có thể tiêu diệt hắn, và lãnh đạo các quốc gia buộc phải tuân theo điều kiện quái gở của con quái vật này: Để hắn làm giáo viên chủ nhiệm cho lớp 3-E của trường Trung học Kunugigaoka. Từ đây đến tháng Ba năm sau, lớp 3-E sẽ có cơ hội ám sát con quái vật này, bảo vệ Trái đất và nhận phần thưởng mười tỷ yên. Tuy nhiên, lớp 3-E lại bị xem là lớp thất bại nhất của trường Kunugigaoka. Những học sinh có vấn đề nhất của trường, những kẻ bị bỏ lại phía sau nay gánh vác trên vai trọng trách của một "Lớp học ám sát".
