Djamari menggantikan Budi Gunawan yang sebelumnya diberhentikan pada reshuffle kabinet, Senin (8/9/2025). Presiden Prabowo Subianto kembali melakukan perombakan kabinet pada Rabu (17/9/2025). Pada reshuffle hari ini, Prabowo melantik dua menteri yang kosong usai perombakan kabinet 8 September 2025 kemarin.
Presiden Prabowo resmi melantik Letjen (Purn) TNI Djamari Chaniago sebagai Menteri Koordinator Bidang Politik dan Keamanan (Menko Polkam) menggantikan Budi Gunawan. Kemudian Presiden Prabowo menggeser Menteri Badan Usaha Milik Negara (BUMN) Erick Thohir sebagai Menteri Pemuda dan Olahraga, sementara itu pengganti Erick Thohir sebagai Menteri BUMN belum diputuskan.
Presiden Prabowo juga melantik Afriansyah Noor menjadi Wakil Menteri Tenaga Kerja, Farida Farichah Wakil Menteri Koperasi, Wakil Menteri Kehutanan Rohmat Marzuki, lalu Angga Raka Prabowo menjadi Kepala Badan Komunikasi Pemerintah.
Naniek S Deyang sebagai Wakil Kepala BGN, Sony Sanjaya sebagai Wakil Kepala BGN, Muhammad Qodari sebagai Kepala Staf Kepresidenan, Sarah Sadiqa sebagai Kepala LKPP, dan Ahmad Dofiri sebagai Utusan Khusus Presiden bidang Kamtibmas.
